Roivant Sciences Ltd. (NASDAQ:ROIV) and its subsidiary Priovant Therapeutics dropped some good news on Wednesday: two Phase 3 trials testing brepocitinib in noninfectious uveitis (NIU) both came back positive. NIU is an inflammatory eye disease that can cause permanent vision loss, so this is not a small deal.
The investigational therapy hit its primary endpoint in both the CLARITY-1 and CLARITY-2 studies. It also showed statistically significant improvements across all key secondary measures, and that held up even with a strict steroid-reduction schedule. In plain English, the drug worked while patients were being weaned off steroids, which is the hard part.
Worth noting: the company's Lisraya (brepocitinib) is already the first and only targeted oral therapy approved for adults with dermatomyositis. So this is not a shot in the dark. It is an established molecule looking for a second act.
Why Noninfectious Uveitis Matters
NIU is a severe inflammatory eye condition and one of the leading drivers of blindness in working-age individuals. Without sufficient treatment, it triggers permanent sight loss along with eye pain, floaters, and extensive visual deficits that affect clarity, color perception, night vision, and peripheral focus.
Those symptoms disrupt essential daily tasks like reading, driving, working, and self-care for roughly 70,000 to 190,000 American adults. That is the patient population sitting in the crosshairs here.
What the CLARITY Trials Actually Showed
Brepocitinib consistently lowered the rate of treatment failure across several patient groups. That covers intermediate, posterior, and panuveitis, as well as participants with or without previous TNFi therapy. The drug also demonstrated efficacy across underlying conditions such as sarcoidosis, birdshot chorioretinopathy, and idiopathic cases.
On the secondary endpoints, the data showed sustained steroid-free quiescence at Week 48, resolved macular edema, improved chorioretinal vascular leakage verified through wide-field fluorescein angiography, and elevated visual functioning scores on the NEI VFQ-25 questionnaire. That is a lot of boxes checked.
Tolerability and the Road to Approval
Patients tolerated brepocitinib well throughout the studies. Most adverse events remained mild to moderate, and adverse events prompted treatment discontinuation in just 3% of trial participants. The trials revealed no new safety signals, which lines up with the drug's established profile.
Priovant plans to submit a supplemental New Drug Application for brepocitinib in NIU during the first half of 2027. That is the next catalyst to watch.
ROIV Price Action: Roivant Sciences shares were up 4.30% at $36.42 at the time of publication on Wednesday, according to market data.
