Enliven Therapeutics (ELVN) got a vote of confidence from William Blair on Tuesday, as the firm kicked off coverage with an Outperform rating. The reason? The company's lead drug candidate, ELVN-001, could shake up how chronic myeloid leukemia (CML) is treated.

ELVN-001 Shows Positive Phase 1 Data
ELVN-001 is a small-molecule kinase inhibitor designed to zero in on the BCR::ABL1 gene fusion, the oncogenic driver of CML. In June, Enliven reported encouraging data from the ongoing ENABLE Phase 1 trial, which is testing the drug in patients with previously treated CML.
The numbers are worth a closer look. In the 80 mg once-daily (QD) Phase 1b cohort, the drug achieved a 61% overall major molecular response (MMR) rate, with 48% of patients hitting MMR by 24 weeks. The response rates were even better in earlier-line patients: among those with only one or two prior unique tyrosine kinase inhibitors (TKIs), 67% achieved overall MMR, and 55% reached MMR by 24 weeks.
Enliven Therapeutics Has Cash Runway Into 2030
As of June 30, 2026, Enliven had $895.2 million in cash, cash equivalents, and marketable securities. That's enough to fund operations into 2030, giving the company plenty of breathing room to advance ELVN-001 through the clinic.
William Blair sees ELVN-001 as a potential treatment for patients across the entire CML continuum, starting with the growing late-line patient population and eventually moving into earlier lines of therapy.
William Blair Sees $8.8 Billion CML Market
Analyst Andy Hsieh pointed out that despite multiple approved TKIs for CML, there's still a significant unmet need. A growing pool of late-line patients is looking for options that are more effective, more tolerable, and more convenient.
William Blair estimates the total addressable market for CML at roughly $8.8 billion, with $5.6 billion of that coming from the U.S. alone.
Hsieh also highlighted a key differentiator: the 48% MMR rate at the planned Phase 3 dose is twice what current market leader Novartis's (NVS) Scemblix (asciminib) has demonstrated. And ELVN-001 is the only TKI that has shown activity against treatment-emergent resistance mutations to Scemblix.
That's a big deal. It means ELVN-001 could step in where Scemblix falls short, potentially disrupting the CML treatment landscape. Hsieh emphasized the drug's differentiated profile and its potential to serve patients across multiple lines of therapy.
As for the stock, Enliven shares were down 3.80% at $1.52 at the time of publication on Tuesday, according to market data.
